组合化学(Combinatorial Chemistry)是用巧妙的方法和步骤,利用某些单体或基本化合物,快速合成大量化合物[1],自此概念的提出以来,已受到药物研究界的广泛关注和探索。但如何从大规模,复杂的化合物库中高效快速的筛选出具有生物活性的物质,是目前医学研究和药物开发面临的巨大挑战。匈牙利罗兰大学Arpad Furka教授在Wiley发行的MedComm上发表展望文章“Combinatorial echnology revitalized by DNA-encoding”, 重点阐述了两种重要的化合物筛选分析方法,OBOC(One Bead One Compound)库和DNA编码组合化合物库(DNA-Encoded Combinatorial Library,DECL),尤其是DECL的原理、优势、发展现状及应用前景,DECL有望快速精准和经济地大量合成和筛选药物,实现药物研究的速度和效率上的巨大飞跃,为将来更多的疾病治疗的新药开发奠定基础[2]。
